چکیده

Background: Bile acids (BAs) affect cellular membranes Results: BAs stabilize domains in plasma membranes, leading to reorganization of membrane proteins and signaling perturbations Conclusion: BAs affect cell function by modulating the stability of plasma membrane nanodomains Significance: These results suggest mechanisms for regulation of functional membrane domains and non-receptor mediated BA signaling

The Bile Acid-Sensitive Ion Channel (BASIC) Is Activated by Alterations of Its Membrane Environment

The bile acid-sensitive ion channel (BASIC) is a member of the DEG/ENaC family of ion channels. Channels of this family are characterized by a common structure, their physiological functions and modes of activation, however, are diverse. Rat BASIC is expressed in brain, liver and intestinal tract and activated by bile acids. The physiological function of BASIC and its mechanism of bile acid act...

Interaction of two bile acids, a cytotoxic and a cytoprotective, with biomembrane models

Bile acids (BA) are naturally-occurring detergents that solubilize dietary lipids in the intestinal tract. However, they are involved in the triggering of diseases like colon cancer and other liver problems. This study has the purpose of investigating how these acids interact with cell membranes and their effects on them. We studied the effects of two bile acids, a cytotoxic (DCA) and other cyt...

Two Distinct Mechanisms for Bilirubin Glucuronide Transport by Rat Bile Canalicular Membrane

Bilirubin is conjugated with glucuronic acid in hepatocytes and subsequently secreted in bile. The major conjugate is bilirubin diglucuronide. Using sealed vesicles which are primarily derived from the canalicular (CMV) and sinusoidal (SMV) membrane vesicle domains of the plasma membrane of hepatocytes, we demonstrated that bilirubin glucuronides are transported byCMV by both ATPand membrane po...

Bile acids modulate signaling by functional perturbation of plasma membrane domains.

Eukaryotic cell membranes are organized into functional lipid and protein domains, the most widely studied being membrane rafts. Although rafts have been associated with numerous plasma membrane functions, the mechanisms by which these domains themselves are regulated remain undefined. Bile acids (BAs), whose primary function is the solubilization of dietary lipids for digestion and absorption,...

Structure-cooperativity relationship in ileal bile acid binding proteins

Background: Ileal bile acid binding proteins display different degrees of binding cooperativity. Results: The structure of a low cooperative protein complexed with bile salts was determined. The protein was mutated to enhance its cooperativity. Conclusion: The cooperative binding requires few latch residues that stabilize an H-bond network. Significance: Knowledge of the determinants of coopera...
